LifeX TruAgeTM determines your Biological age by measuring DNA methylation levels in specific DNA regions. Scientists at LifeX TruAgeTM focus on analyzing the methylation of cytosine residues, particularly in CPG islands—small DNA regions clustered around genes that influence gene activity. This methylation, performed by DNA methylases, is linked to gene inactivation, playing a crucial role in cancer development and cell aging. Reversing DNA methylation at these sites is considered a potential therapeutic strategy to restore gene expression and enhance physiological function. Bisulfite conversion is utilized to determine the methylation status of a DNA sequence. This process converts unmethylated cytosines into uracil while leaving methylated cytosines unaffected. By sequencing 9 small regions of your DNA and assessing the degree of methylation, our algorithm calculates your true biological age. It's important to note that we don't sequence your entire genome, and we don't retain your saliva sample or DNA; all biological material and DNA sequence data are deleted upon completion of the test.
Upon receiving your sample at LifeX TruAgeTM we begin the process by scanning its identification number into our internal registration system. The initial step involves a visual inspection to ensure the sample is free from contamination and falls within the appropriate moisture range. If a sample doesn't meet these criteria, we promptly notify the customer and provide a replacement kit for re-collection. For samples that pass the visual inspection, the subsequent stages include DNA extraction and bisulfite conversion to distinguish between methylated and unmethylated DNA. Following this, small regions of your DNA undergo extraction and sequencing to quantify the percentage of DNA methylation. All sequencing procedures are conducted by independent laboratory services. Once the analysis is successfully completed, the laboratory sends the resulting data, along with the sample's barcode, to LifeX TruAgeTM. This enables us to match the information with the correct account and initiate the interpretation of your data. It's worth noting that while LifeX TruAgeTM can calculate your biological age without needing your date of birth, providing additional details such as your date of birth, lifestyle, general health status, and race helps us fine-tune our algorithm and improve our services.

The accuracy of the LifeX TruAgeTM may be compromised in instances where an insufficient amount of DNA is isolated from your sample or if the sample is contaminated. Contamination typically occurs when the sample isn't adequately dried, leading to bacterial or mold overgrowth on the filter paper.
